Intrinsic Morphologic and Physiologic Development of Human Derived Retinal Ganglion Cells In Vitro.

This study examined the intrinsic morphologic and physiologic development of human retinal ganglion cells. Cells were derived from human pluripotent stem cells and cultured for 7 weeks. The cells continued to mature over this time. The maturation state of the cells should be considered when considering their use as treatments for optic neuropathies.

Collagen Mimetic Peptides Promote Corneal Epithelial Cell Regeneration.

This study investigated the capacity of a collagen mimetic peptide (CMP) to promote corneal epithelial healing after an acute wound. A 360o lamellar keratectomy was performed and wound closure was compared between treated and vehicle groups. Treatment with CMP increased adherence of the basal epithelium to the underlying substrate and enhanced density of epithelial cells. This may represent a novel therapeutic to heal corneal tissue by repairing underlying collagen after corneal injury.

Effects of intravitreal injection of siRNA against caspase-2 on retinal and optic nerve degeneration in air blast induced ocular trauma.

Repeat exposure to over-pressure air causes optic nerve damage. Treatment with caspase-2 siRNA was protective, but only when given prior to injury. Notably, intravitreal injections given soon after injury increased intraocular inflammation, which counteracted the positive effect of caspase 2 siRNA.

This is a review of the evidence on safety and efficacy of anti-VEGF and intravitreal corticosteroid pharmacotherapies for the treatment of macular edema (DME). The published literature indicates that intravitreal injections of anti-VEGF agents and corticosteroids are efficacious treatments for DME.

The goal of this study was to understand mechanisms underlying differences in efficacy and toxicity between intra-arterial chemotherapy and intravenous chemotherapy. Greater efficacy and lower systemic toxicity was associated with intra-arterial chemotherapy. This appeared to be attributable to the greater ocular-to-systemic drug concentration ratio compared with intravenous chemotherapy.

Racial/Ethnic Disparities in Ophthalmology Clinical Trials Resulting in US Food and Drug Administration Drug Approvals From 2000 to 2020.

In this cohort study, Black, Hispanic or Latinx, and other non-White participants were underrepresented in clinical trials leading to FDA ophthalmology drug approvals compared with the expected disease burden and racial/ethnic distribution in the US. Although there was meaningful improvement from 2000 to 2020, further efforts to increase minority enrollment in clinical trials seem to be warranted.

Insect Foreign Body in a Boston Type I Keratoprosthesis.

Clinical case report of a corneal transplant patient who presented for eye pain and foreign body sensation. She had a history of melting corneal ulcer requiring multiple corneal transplants and eventual Boston keratoprosthesis. The foreign bodies were removed without complication.

Retinal Artery Occlusion After a Dog Bite in a 55-Year-Old Man.

Clinical case report of unilateral uveitis and 2 days of sudden vision loss along with self-reported light sensitivity and mild eye pain. An inferior hemicentral retinal artery occlusion was confirmed on intravenous fluorescein angiography. Notably, a dog bit the patient’s hand two months prior to presentation and was associated with a persistent cough, night sweats, and low-grade fevers despite two courses of antibiotics.

We found WldS protects dendritic morphology and light-evoked responses of RGCs that signal light onset, but not those that respond to light offset during IOP elevation. As expected, WldS preserves anterograde axon transport and spatial acuity in young adult mice, but its protection is significantly limited in aged mice.

Retinal oxidative stress activates the NRF2/ARE pathway: An early endogenous protective response to ocular hypertension.

We found that the NRF2-KEAP1-ARE pathway is endogenously activated early in ocular hypertension due to phosphorylation of NRF2 by the PI3K/AKT pathway. Further, activation of this pathway slows the onset of axon degeneration and vision loss after IOP elevation. Exogenous activation of this pathway might be an effective neuroprotective approach.

Angiographic investigation of orbital vascular variations in the rabbit and implications for endovascular intra-arterial chemotherapy models.

We show that successful catheterization of the ophthalmic arteries in the rabbit can be readily accomplished with nuanced technique. The external ophthalmic artery is the dominant arterial supply in the majority of rabbit eyes. These findings allow for successful reproduction of ophthalmic artery catheterization studies of intra-arterial chemotherapy for retinoblastoma in rabbits.

Human pluripotent stem cells were chemically differentiated into hRGCs and cultured up to 7 weeks. We measured soma area, neurite complexity, synaptic protein, axon-related messenger RNA and protein, and voltage-dependent responses. We found that human RGCs develop up to 7 weeks after culture. Thus, the state of hRGC maturation should be accounted for in designing models and treatments for optic neuropathies.

Chronic low-grade retinal inflammation is an essential contributor to the pathogenesis of diabetic retinopathy (DR). We investigated the NFAT-dependency of IL-1β-induced inflammation in human Müller cells (hMC) and human retinal microvascular endothelial cells (hRMEC). Our data demonstrate the potential of NFAT inhibition to mitigate retinal inflammation secondary to diabetes.

Altered Ocular Fibrillin Microfibril Composition in Mice With a Glaucoma-Causing Mutation of Adamts10.

Previously, we identified a G661R mutation of ADAMTS10 (a disintegrin-like and metalloprotease with thrombospondin type 1 motif 10) as being disease causative in a colony of Beagles with inherited primary open-angle glaucoma (POAG). Here we show that Adamts10G661R/G661R mice recapitulate the short stature and ocular phenotypes of Weill-Marchesani syndrome (WMS). The altered fibrillin-1 and fibrillin-2 immunoactivity in Adamts10G661R/G661R mice suggests that the G661R mutation of Adamts10 perturbs regulation of the fibrillin isotype composition of microfibrils in the mouse eye.
